The third case involved a man in his 50s with scleroderma who died in March 2026 after receiving an experimental in-vivo CAR-T therapy developed by RiboX Therapeutics.
RiboX had announced US Food and Drug Administration clearance to begin testing the treatment in the United States shortly before the death became public. Public reports have not established that the therapy caused his death, making further investigation important before conclusions are drawn.

Gene and Cell Therapies Can Trigger Serious Reactions
Gene therapies use delivery systems that often modified viruses to carry genetic material into a patient’s cells. Although these vectors are engineered for medical use, the immune system may still recognise them as foreign and produce a powerful inflammatory response.
High doses may increase this risk because they expose the body to a larger quantity of the vector. In the Duchenne muscular dystrophy case, the child reportedly developed acute respiratory distress syndrome during a severe immune reaction.
Acute respiratory distress syndrome causes inflammation and fluid accumulation in the lungs, severely limiting the transfer of oxygen into the bloodstream. It can progress rapidly and become life-threatening.
